Phase: 1/2
Start date: May 12, 2025
Planned enrollment: 130
EIK1004 (formerly IMP1707) is an investigational cancer drug; EIK1004-001 is its ongoing phase 1/2 trial, not another name for the drug. The trial is testing EIK1004 alone in people with advanced ovarian, breast, prostate, or pancreatic cancer and selected DNA-repair gene mutations, including some people with brain metastases. (aacrjournals.org)
EIK1004 is designed to inhibit PARP1, a DNA-repair enzyme, while largely sparing PARP2. In laboratory tests it also trapped PARP1 on DNA; in mouse models it reached the brain and caused regression of tumors implanted there. These are preclinical findings, not evidence of benefit in people. (aacrjournals.org)
Limited human results have been disclosed, but no reliable response rate is available. In a company filing reporting data through October 27, 2025, 16 patients had received doses of 10–40 mg, and one unconfirmed partial response was reported at 10 mg. The filing did not establish how many patients were evaluable for response. The company planned to present its first fuller clinical dataset in October 2026. (sec.gov)
The same filing reported no dose-limiting toxicities among the 16 enrolled patients as of October 27, 2025. That small, early dataset cannot establish the drug’s overall safety profile. (sec.gov)

Goal: Determine a tolerable dose and characterize the safety, pharmacology, and preliminary antitumor activity of EIK1004 monotherapy in advanced solid tumors with selected homologous recombination repair (HRR) gene mutations.
Patients: Adults with recurrent advanced or metastatic breast, ovarian, castration-resistant prostate, or pancreatic cancer and deleterious or suspected deleterious germline or somatic mutations in selected HRR genes. Patients need ECOG performance status 0–1, adequate organ function, and prior treatment appropriate to their cancer type. Selected patients with CNS metastases may enroll; prior PARP inhibitor exposure is limited to one line, and prior PARP1-selective inhibitors are excluded.
Design: Open-label, multicenter, nonrandomized dose-escalation and dose-optimization study. Part 1 seeks the maximum tolerated or maximum achievable dose; Part 2 evaluates selected doses further.
Treatments: EIK1004 (IMP1707) is given as oral monotherapy, generally daily after a single-dose period, until progression or discontinuation. It is an investigational PARP1-selective inhibitor designed to impair DNA repair and trap PARP1 on DNA while largely sparing PARP2. In a preliminary company report of 16 treated patients, there were no dose-limiting toxicities and one unconfirmed partial response; these data are insufficient to establish efficacy or overall safety.
Outcomes: Primary measures are dose-limiting toxicities during the first 28 days and adverse events, including serious events, through one month after the last dose. Secondary measures include plasma pharmacokinetics and objective response assessed using disease-appropriate criteria, including RECIST 1.1, RANO-BM, CA-125, or PSA. Plasma cytokine changes are also assessed.
Burden on patient: Likely high, particularly during dose escalation: daily oral treatment is accompanied by close toxicity monitoring, ECGs, laboratory testing, pharmacokinetic blood draws, and response assessments, requiring more clinic visits than routine care. The supplied information does not specify visit frequency or mandatory biopsies.
